Barrier properties of varnishes for protection of photographs

In this diploma thesis the basic principles of inkjet print, ink composition, print media and degradation mechanisms of ink dyes are summarized. The thesis deals with study of varnish barrier properties in coloured inkjet prints. Permeability of water vapour and oxygen (parts of atmosphere) depending on different coatings are tested there. In addition the influence of ozone on degradation of dyes was studied, while ozone was permeating through barrier varnish layer of inkjet print. Ozone plays an important role in degradation processes of the inkjet prints and it also causes a gas-fading. Concentrations of acting ozone were established by iodometric titration. Simultaneously kinetics of degradation of Orange 7 was studied on indicator papers.
Isolation, characterization and application of biomedically important polymer P(3HB-co-4HB)

This diploma thesis deals with the isolation and characterization of copolymer P(3HB-co-4HB). The teoretical part was prepared as a literature search which describe polyhydroxyalkanoates in general, their structure, synthesis, degradation and isolation. Furthermore, copolymers containing 4HB units are mentioned in this thesis and there is also no mentioned of the biosynthesis and biodegradation of copolymer P(3HB-co-4HB). The first aim of this diploma thesis was to find out which solvent is the best for copolymer extraction and then characterize obtained copolymer P(3HB-co-4HB). The isolated copolymer was characterized by gas chromatography, SEC-MALS, thermal analysis and SEM. In the second part of the thesis, release of model biologically active substance from the PHA films was studied. Rhodamine 6G dye was selected for the simulation, which was used to stain the copolymer films and the P(3HB) films. It was observed that film prepared from P(3HB-co-4HB) copolymer released entrapped substance much faster than film made from P3HB homopolymer, and, in addition, the copolymer was substantially more susceptible to enzyme degradation.

This thesis is focused on diffusion of dyes in biopolymer-based hydrogels. These hydrogels are based on interaction between biopolymer-like electrolytes with oppositely charged surfactants. When polyelectrolytes interact with oppositely charged surfactants, micelle-like nano-containers can be formed. These nano-containers are able of binding hydrophobic compounds. In this study, combination of modified dextran (diethylaminoethyl dextran) with positive charge and oppositely charged sodium dodecylsulphate as surfactant was used for preparation of hydrogels. Next type of hydrogel was based on hyaluronan and positive charged surfactant Septonex (carbethoxypendecinium bromide). As a diffusion probes in hydrogels dyes Nile red and ATTO 488 were used. The diffusion of these dyes from aqeous solutions of NaCl or surfactants into hydrogels was monitored in time. Transport of dyes into structure of hydrogels was characterized by diffusion coefficients and structural parameters of hydrogels.
New Approaches in Cardiac Optical Mapping

This dissertation deals with new approaches in cardiac optical mapping. The principle of cardiac optical mapping as well as the current research in this field was described. The new measurement system was developed and its characteristics presented. The system design allows epicardial and endocardial mapping, employing new ratiometric techniques in 2D acquisition and simultaneous electrical and optical mapping. The measured characteristics of fluorescent dye di-4-ANEPPSS were presented. The relation between movement and consequent motion artifacts is well described. Several new approaches in signal processing were proposed, including new ratiometry technique and using image registration to suppress motion artifacts. The algorithm for elastic image registration of optical signals and innovative method for verification of registration process were presented. Application of all proposed approaches and its results are included and discussed.

Title of Diploma Thesis: Multicomponent analysis This diploma thesis deals with the determination of concentrations of individual dyes in mixtures by multicomponent spectrophotometric analysis. The method was tested on selected dyes which were divided into two groups. The first group contains methyl orange, methyl red, orange II and phenol red. The second group consists of methylene blue, bromocresol green and thymol blue. Within each group mixtures were prepared containing combinations of two, three and four dyes in various ratio. Spectra of prepared mixtures were measured by diode array UV/VIS spectrophotometer. Absorbance values were read at wavelengths of absorption maxima of the individual dyes. Using the pseudoinverse matrix, concentration ratios of components were calculated from the absorbance values. The determination in determined and over determined systems was performed. On the basis of the results obtained by the method of analysis it is possible to determine the individual concentration of the multi-component dyes mixtures without prior separation. Colorants in medicinal products and their influence on health

The aim of this diploma thesis was to create an overview of colorantsauthorized for use in pharmaceutical products, conditions of their use and to characterize selected colorants in detail. The thesis is focused on the analysis on the health impact of the colorants. Their connection with child behaviour disorders and allergic reactions is described minutely. Part of the thesis is devoted to pointong out which colorants show the least negative impact on health as well as colorants with the highest incidence of side effects. A chapter describing the legislation on colorants in pharmaceuticals is also included. It is obvious, that natural colorants present a lower risk of side effects than synthetic colorants. Synthetic colorants with the highest incidence of side effects include the richly represented group of azo colorants, which causes ADHD in children and in hypersensitive patients, along with the natural colorant carmine, also allergy. Porovnání anthokyanů u modrých odrůd

This thesis deals with the comparison of anthocyanins in blue grape varieties. It has been compared 23 varieties of Vitis vinifera L. and 9 interspecific varieties from the harvest 2015. The grapes come from the wine region Mikulov and from the wine village Lednice. Literary part deals with the chemical structure of anthocyanins and their occurrence in grapes, further accumulation of anthocyanins and influencing factors, physico-chemical factors affecting the stability of anthocyanins, change of anthocyanins during the vinification and extraction of anthocyanins and measurement of color parameters. The practical part is especially focused on compare the content of anthocyanins and their profile in varieties of Vitis vinifera L. and interspecific varieties.
